New, High-performance Solid Support for Oligonucleotide Synthesis Developed

Nitto Denko Makes Another Stride Forward in Biomedical Field

Japan's leading diversified materials manufacturer Nitto Denko Corporation (Nitto Denko) and its wholly owned U.S. research subsidiary Nitto Denko Technical Corporation (NDT) [Nitto Denko and NDT hereafter collectively called “Nitto Denko Group”], have jointly developed a new, high-performance solid polymer support for oligonucleotide synthesis, Nitto Denko announced today.

Nitto Denko Group is currently marketing solid support used for chemical synthesis of nucleic acid drugs such as DNA and RNA, under the trade name “NittoPhase®”, via another fully owned U.S. subsidiary of Nitto Denko, Kinovate Life Sciences Inc. (KLS).

The recently developed solid support can be loaded at significantly higher levels when compared to existing products on the market, making it possible to synthesize nucleic acids at with high purity and high yield. Furthermore, the new solid support has demonstrated high performance in the synthesis of RNA which have until now been more difficult to synthesize.


Background of the Latest Development

Nucleic acid drugs are a relatively new class of molecules that have in the past few years shown good promise for the treatment of various illnesses through the delivery of oligonucleotides such as antisense DNA and RNA into the human body. In general, short DNA and RNA oligomers are chemically synthesized on the surface of a solid support, and are then used as nucleic acid drugs after cleavage from the support and subsequent purification.

Initial research into nucleic acid drugs focused on antisense DNA, consisting of single strand DNA oligomers. However, in recent years “siRNA (small interfering RNA) drugs” comprising oligomers of double strand RNA have attracted much attention. As a result, a further expansion of the synthesis market for nucleic acid drugs, including siRNA drugs, is expected in the future.

It is with this backdrop that Nitto Denko Group has developed the improved version of the existing NittoPhase® solid support, which is capable of loading up to the highest level in the world, which enables the synthesis of not only DNA but also RNA, with high yield as well as high purity.

Product Characteristics

Owing to the unique polymer design elements such as chemical composition and physical structure, the new solid support has extremely high loading capacity, enabling the synthesis of oligonucleotides such as DNA, as well as RNA which have been traditionally more difficult to synthesize, not only at with high yield and purity.

Business Development Going Forward

Nitto Denko Group is scheduled to introduce the new solid support at the TIDES(*) Meeting to be held at Las Vegas, U.S.A. from May 17 to 20, 2009, and to start distributing samples via KLS. In future, Nitto Denko Group plans to expand further into other areas of the nucleic acid synthesis related market.
